Other justification : CosMx® Spatial Molecular Imager (SMI) is a novel in situ technology (proprietary to Bruker Spatial Biology) that combines the power of high-plex profiling with high-resolution imaging and allows researchers to visualize and quantify targeted protein and gene expression on formalin-fixed paraffin-embedded (FFPE) tissue, fresh frozen (FF) tissue, tissue microarrays (TMAs) or organoids on a microscope slide. In contrast to most current single-cell analysis technologies, which are applied to dissociated cells, CosMx SMI enables researchers to analyze hundreds to thousands of RNAs or proteins directly from single cells, down to subcellular resolution, within morphologically intact tissue samples. This provides a higher multiplexing capacity (6,000+ targets) all with spatial context from formalin-fixed, paraffin-embedded (FFPE) or fresh frozen (FF) tissue sections. The CosMx SMI platform is an integrated system with mature cyclic in situ hybridization chemistry, ultra-high-resolution imaging readout instrument, and cloud-based, interactive data analysis and visualization software. Sample preparation involves standard in situ hybridization (ISH) processing steps and allows direct use of pathology lab-standard glass slides without any tissue expansion or clearing and does not require any cDNA synthesis or amplification. RNA or protein targets in individual cells are identified through hybridization of targets with specific probes or antibodies labelled with a unique barcode system, followed by readout of the barcode with cyclic rounds of fluorescently labeled reporter probe imaging. Each RNA appears as a single spot in cells and digitally quantified by counting the number of imaged spots. All data is then migrated to cloud storage where users will be provided with analysis and visualization tools and will also have the opportunity to incorporate custom analysis workflows.
