Title : ID7000Cell Analyzer
Description : The ID7000 spectral flow cytometer allows for a detailed high-dimentional analysis of single cells from a homogenous suspension. This can be from cell cultures or clinical and pre-clinical tissue samples. Based on simultaneous immunophenotyping of surface and intracellularly expressed proteins this will enable us to characterize the cell types, subpopulation-distribution and cell purity in our samples, which will be important in the work with EVs in diagnostics, vaccines and cancer research. Our current options with regular filter-gated flow cytometers only enable us to use 13 detectors, which gives issues covering the full complexity of the samples based on state-of-the-art knowledge. The ID7000 six-laser system will allow us to use 182 detectors, thereby greatly enhancing our current options. The total of 182 detectors will enable us to do an in-depth analysis of the cell populations in our tissue samples. Further, more available lasers and detectors will give us a much greater flexibility in selecting fluorochromes for the panels. Further it will enable all markers to be run in a single panel, which will increase the amount of information gained from each sample. Compared to running multiple parallel panels this will reduce the overall running cost and minimize the use of patient material needed for analysis. Including a 320 nm UV laser will enable analysis of samples stained with DAPI or Hoechst DNA stains enabling high-resolution analysis of cell cycle phases, cellular DNA characteristics and DNA polity. The ID7000 is the only current spectral flow cytometer that include six lasers with a 320 nm option. Further, the ID7000 is the only current spectral flow cytometer that include 182 detectors. Current regular flow cytometry requires compensation to separate spectral overlap and requires subjective end-analysis. The ID7000 uses a spectral unmixing algorithm to separate complex fluorescence signals, eliminating the need for compensation, and the nature and complexity of the data allowing un supervised analysis using dimension-reduction algorithms like tSNE. The ID7000 has a fully integrated AutoSampler that supports true walk-away operation. The system can be programmed for automatic shutdown after a run. A fully automated system will reduce the hands-on time from technical staff and researchers. Another important aspect of the ID7000 is the free analysis Software which can be downloaded as many times as required by all end users. This is important since several users from hospital and university will be using the instrument. For future purpose, the 6-laser optical capability is matched to Sony's FP7000 Spectral Cell Sorter thus enabling multicolor assay portability from the cell analyzer to the cell sorter. We aim at acquiring the unique FP7000 Spectral Cell Sorter in the future, and the compatibility between the two systems will save time and money during panel design and optimization. In conclusion: The ID7000 system is the only current instrument that meet our requirements for number of lasers, detectors and laser composition and will enable future transition between the analyzer instrument and a sorting option (FP7000).
